Yes. If you have two supplies available on the SBC,
you can power the output buffers from 5V and drive
5V levels, and you can power the input buffers
from 3.3V and receive 5V logic levels.

I tend to put source termination resistors on the
outputs of the buffers, so that I can tweak things
if needed, and then put 100-Ohms in series with
any headers and the buffer inputs, just as a
token attempt at some ESD protection. So basically
an R on the input and output of each buffer on
your test board would give you a little extra
functionality if needed.
