It is my understanding that you can't boot from a thumb drive.
TS has code to make possible boot from on board flash, or SD card.
I think the issue is the bootloader. The bootloader must know how to mount the filesystem.
Apparently the bootloader does not know about thumb drive file system.

Of course, you can boot a kernel elsewhere, then mount the thumb drive as root.
