As a game developer who works on the 360 and PS3, I can tell you that
sampling profilers such as this most certainly DO work and can be
quite a valuable tool in determining what routines may need
optimization.  I can also confirm that the CPU penalty for such a
profiler is VERY low compared to an instrumenting profiler(which I
also use).  This is good news if his interrupt handler stuff actually
works on our platform.
